4L80E.. The real deal..
So I am doing a 5.7 LS1 swap in a 63 Chevy pick up. I have:
Early 4L80E ( I know one bolt hole won't line up)
Stock 4L80E Converter (but will be buying a 2400rpm Yank verter for it)
4L80E Flex plate
99-02 F-body LS1 pcm / Harness but have bought the 4L80E connectors and pins from Speartech
HP tuners unlimited 99-02
I know I'm going to need:
Custom drive shaft
Custom Trans mount
VSS Sensor
Will I need anything like spacers between the flex plate and verter?
What will I need for a shifter? I was thinking any TH350/400 shifter would work, just will only have a lock in 2nd, 3rd , 4rth if manually shifted.
What am I not thinking of?

you'll need the spacer between the block and flywheel, not between the converter, unless your 4L80E flexplate has one pressed in it already (mine did). I have one, PM me if you need it. Other than that I think you've got it covered.

Yes, the spacer goes on the end of the crank and then you put a 6.0L flexplate on. A standard 5.3L flexplate will not work with the spacer. There are also longer bolts that you should get from GM for the 6.0L/crank spacer combo to bolt to the crank flange.
Any shifter setup for a 4speed auto(700r4, 4L60E or 80E) will work. I don't know about the 350/400 shifters but, you could always give it a shot if you have one laying around.

Doesn't the '01 and up 6.0L flexplate have the spacer built into it??? My 6.0 came out of a truck with a 4L80E, and I swapped out my stock 5.3 flexplate when installing it. I'm gonna be using the 6.0 flexplate with my 4L80E swap, with no spacer.

Yeah, the '01 up 6.0L(80E trucks) plates have the spacer already pressed on the flexplate because the crank flange is shorter than the previous year 6.0's.
What year 6.0L do you have Tim?

There was No way the spacer would fit between the crank and flex plate, and still allow the starter to engage.
BUT this is 5.3 flex plate with a 4l80e converter with 5.3 mounts....
The only reason for the spacer was to support the Torque converter.
