E85 will be your best friend if you go with the turbo. Just gives you more of a reason to pull the trigger.
You will want to measure converter slip going down the track. You don't want it slipping 20% when you're in the throttle. It will be slow and cause a **** ton of heat. You'll probably need to a specd converter for your truck once you get rolling.

Ive run up to 17 psi on my 10.6:1 lq9 on E85 and it's still kicking no problems. Only major problem I have is don't think the stock fuel lines can support that type of HP. My second walbro 450 is kicking on but it's not helping. I've been too lazy to add -8 lines and regulator.
E85 is the ticket but I'm getting a bit sick of it just because there's only 1 freaking station anywhere close to me. Houston is massive yet there's only a few stations that sell it in Houston proper.

Ive got some 127 lb injectors for sale if you're interested.
Ive run up to 17 psi on my 10.6:1 lq9 on E85 and it's still kicking no problems. Only major problem I have is don't think the stock fuel lines can support that type of HP. My second walbro 450 is kicking on but it's not helping. I've been too lazy to add -8 lines and regulator.

I think it will get there or be pretty damn close. High compression LS3 headed 6.0L and a 78mm turbo. I think he's got it in the bag.
I made 578 with my set up on 12psi. Hell, vicalma03 put down 650rwhp on 13psi with his 4.8L and 80mm compound set up. The OP's compression will bring those power numbers up on low boost settings.
