I basically want to know, how to make it work. My choices right now are the 6l80 or 90, 4l60 or 4l65. I have a local friend who has his t-56, i really dont like stick but 6 speed auto would be nice but how complicated is it to make it work behind my application and can it handle the power my motor might make. Currently I have the motor minus the wiring harness and computer, so before I go to Speartech or Street and Performance, I need to know which way to go on the trannies and computer.

What vehicle are you putting this in? For a drop-in application you need a car that uses CAN data. Until a standalone controller is made available, and you want to retro fit a 6L80 in a non CAN car, you will need a E38 PCM or equivalent so the TCM in the 6L80/90 communicates with the engine. You will need electronic throttle also. Speartech can hook you up with the harness.
The 6L80 will fit in any space the 4L60 originally occupied.
Of course there are other things like a new driveshaft, etc but those are easy.

It originally came with an anemic 4l60 but with no CAN as you call it.
I do not want an electric throttle body because i just paid 400 for a nick williams billet 90mm throttle body plus i dont trust the electronic one IMHO. Looks like more parts to go wrong for more money, i'll stick with the cable. Is there anyway to do cable driven and the 6l80e?

Not at this time. Eventually there will be stand-alone controllers that will allow you to operate the 6L80E with cable throttle or even behind a carb motor. A stand-alone was supposed to be available by December 07 but they must be having some issues.

This is the first stand alone device I have seen trinitytransmission.com/6l80e Maybe you can be the beta tester.
