If I were in your shoes i'd probably do a 4L80, while a T56 would be sweet the clutch would worry me + i'm lazy and hate shifting. My buddy has a 6.0 4L80E in his 06 2500 sierra, it's a heavy pig CCLB 4x4. He has towed 20K lbs+ with it. with 60K on the clock the thing doesn't miss a beat, it's a stout trans for sure.

Anybody know about the durability of the 6 speeds? If it were like the 80e it would be awesome. My work truck is an 02 2500hd that has almost 300k on it with the original drivetrain, and **** poor maintainance, but runs like new and pulls a 28' trailor almost every day of its life.
